目的:探究鳕鱼皮胶原蛋白肽对小鼠慢性肝损伤的保护作用机制,为肝病患者功能食品的开发提供理论和试验依据。方法:采用腹腔注射CCl4建立小鼠慢性肝损伤模型,检测鳕鱼皮胶原蛋白肽对小鼠血清中转氨酶和肝组织匀浆中抗氧化酶以及丙二醛的影响;HE染色观察小鼠肝组织病理学变化;蛋白印迹法测定肝组织Bax,Bcl-2,Cleavage Caspase-3,Caspase-9,Cytosolic Cyt c,TNF-α,JNK,P-JNK蛋白的表达情况。结果:与模型组比较,高剂量组(400 mg/kg)小鼠血清中AST、ALT活性、肝组织匀浆中MDA含量显著降低,而肝组织匀浆中SOD活性显著升高,且有显著的剂量关系;小鼠肝组织损伤情况得到明显改善;Bcl-2蛋白表达量增加,Bax,Cleavage Caspase-3,Caspase-9,Cytosolic Cyt c,TNF-α,P-JNK蛋白表达量降低。结论:鳕鱼皮胶原蛋白肽可增强慢性肝损伤小鼠的抗氧化活性,抑制氧化应激,对慢性肝损伤有较好的保护作用。
Objective: To investigate the protective mechanism of cod skin collagen peptide on chronic liver injury in mice and provide theoretical and experimental basis for the development of functional food in patients with liver disease. Methods: The model of chronic liver injury was established by intraperitoneal injection of CCl4, and the effects of cod skin collagen peptide on anti-oxidase and malondialdehyde in transaminase and liver homogenate of mice were detected. HE staining was used to observe the changes of liver tissue The changes of Bax, Bcl-2, Cleavage Caspase-3, Caspase-9, Cytosolic Cyt c, TNF-α, JNK and P-JNK in liver tissue were detected by Western blotting. Results: Compared with the model group, the levels of AST and ALT in the serum of the high-dose group (400 mg / kg) and the content of MDA in the homogenate of the liver tissue were significantly decreased, while the activity of SOD in the homogenate of the liver tissue was significantly increased (P <0.05). The damage of liver tissue in mice was improved obviously. The expression of Bcl-2 protein was increased and the protein expression of Bax, Cleavage Caspase-3, Caspase-9, Cytosolic Cyt c, TNF- α and P-JNK were decreased. Conclusion: Cod skin collagen peptide can enhance the antioxidant activity of chronic liver injury mice, inhibit oxidative stress, and have a good protective effect on chronic liver injury.
