A modified differential display reverse transcription-PCR technique was used to obtain a differentially expressed cDNA fragment induced by oxidized low density lipoprotein from vascular endothelial cells. Using this fragment as a probe, a new full-length cDNA was screened from a human aortic cDNA library. The nucleotide length of 4137bp, encoding a protein of 642 amino acids. Northern blot analysis confirmed that the gene expression was reduced by a factor of 3 when oxidized low-density lipoprotein induced endothelial cells and the gene was designated OLRG-1. OLRG-1 is a multi-tissue expressed gene that is highly homologous to the human NS1-BP gene. Oxidized low-density lipoprotein may regulate gene expression by decreasing the expression of OLRG-1, affecting the splicing maturation of mRNA precursor.
