目的:通过用秋水仙碱给大鼠灌胃复制雄性大鼠生精功能减退模型。方法:20只雄性性成熟大鼠随机分对照组、环磷酰胺组、秋水仙碱低剂量组和秋水仙碱高剂量组,秋水仙碱片以蒸馏水溶解,低剂量组按1.6mg/kg、高剂量组按3.2mg/kg灌胃,每日1次,连续5周;环磷酰胺粉剂则以生理盐水溶解配成20mg/mL浓度,用时按50mg/kg腹腔注射,每周2次,连续5周。对照组方法同上,腹腔注射等量生理盐水(2.5mL/kg)。结果:与对照组比较,秋水仙碱高剂量和低剂量组大鼠睾丸有不同程度的生精小管萎缩,生精细胞减少,排列松散,生精小管间腔隙扩大,管腔内精子细胞数量减少。结论:用秋水仙碱灌胃可复制雄性大鼠睾丸生精功能减退模型。
OBJECTIVE: To make a model of dysfunctional spermatogenesis in male rats by gavage with colchicine. Methods: Twenty male adult rats were randomly divided into control group, cyclophosphamide group, colchicine low-dose group and colchicine high-dose group. Colchicine tablets were dissolved in distilled water. The low dose group was treated with 1.6 mg / kg, High-dose group by 3.2mg / kg gavage once a day for 5 weeks; cyclophosphamide powder was dissolved in saline to 20mg / mL concentration, when used by 50mg / kg intraperitoneal injection twice a week, continuous 5 weeks. The same method as the control group, intraperitoneal injection of saline (2.5mL / kg). Results: Compared with the control group, the testicular achondroplasia of rats in high dose and low dose groups of colchicine reduced, the number of spermatogenic cells decreased, the arrangement was loose, the space between seminiferous tubules enlarged, the number of sperm cells in the lumen cut back. Conclusion: The model of testicular hypofunction can be duplicated with colchicine.
