目的检测原发性肝细胞癌中EphA2的mRNA和蛋白的表达及临床意义。方法应用逆转录-聚合酶链反应(RT-PCR)技术及免疫组织化学方法检测EphA2的mRNA及蛋白在40 例肝癌及相应的癌旁肝组织和10例正常肝脏组织中的表达,并分析其与临床病理特点之间的关系。结果半定量分析显示EphA2 mRNA的表达在肝癌组织(0.836 8±0.205 7)、癌旁肝组织 (0.768 9±0.118 0)和正常肝组织(0.720 8±0.091 0)中的表达差异无统计学意义(P>0.05);从正常组织、癌旁肝组织到肝癌组织,EphA2蛋白的阳性表达率分别为50.0%、57.5%和87.5%。呈递增趋势(X~2=20.21,P<0.01);EphA2蛋白的过表达与肝癌患者的门脉癌栓、卫星灶及淋巴结转移等临床病理因素有关(P<0.05)。结论EphA2蛋白的过表达与肝癌的淋巴结转移、卫星灶的形成和门脉癌栓有密切的联系,提示其在肝癌的侵袭和转移中发挥重要的作用。
Objective To detect the expression of EphA2 mRNA and protein in primary hepatocellular carcinoma and its clinical significance. Methods The mRNA and protein levels of EphA2 were detected by reverse transcription polymerase chain reaction (RT-PCR) and immunohistochemistry in 40 cases of HCC and corresponding paracancerous liver tissues and 10 cases of normal liver tissues. And the relationship between clinicopathological features. Results Semi-quantitative analysis showed that the expression of EphA2 mRNA was significantly increased in HCC tissues (0.836 8 ± 0.205 7), paracancer liver tissues (0.768 9 ± 0.118 0) and normal liver tissues (0.720 8 ± 0 (P> 0.05). The positive expression rates of EphA2 protein in normal tissues, adjacent liver tissues and hepatocellular carcinoma tissues were 50.0% and 57.5%, respectively 87.5%. (P <0.01). The over-expression of EphA2 protein was associated with clinicopathological factors such as portal vein tumor thrombus, satellite tumor and lymph node metastasis in patients with hepatocellular carcinoma (X 2 = 20.21, P <0.01) . Conclusion The overexpression of EphA2 protein is closely related to the lymph node metastasis, the formation of satellite lesions and portal vein tumor thrombus in HCC, suggesting that EphA2 may play an important role in the invasion and metastasis of HCC.
