目的:探讨前列腺素E2(PGE2)和白细胞介素1-β(IL1-β)在单一和联合应用时,对体外培养兔子宫韧带成纤维细胞增殖的影响。方法:第3代幼兔子宫韧带成纤维细胞,以PGE2(25、50、100、500、1 000ng/ml)、IL1-β(10、20、50、100、200 ng/ml)的单独和联合刺激组为实验组,以常规培养组为对照组,通过MTT法测定细胞增殖效应。结果:单独应用时,100 ng/ml的PGE2和20 ng/ml的IL1-β分别为各自最有效的促增殖浓度(P<0.01);以此浓度联合刺激条件下的细胞增殖活性与对照组及单独刺激相比均有升高(P<0.05)。结论:适当浓度内的PGE2与IL1-β单独或联合使用,能有效促进体外子宫韧带成纤维细胞的增殖,说明PGE2与IL1-β对子宫韧带成纤维细胞的增殖有调节作用。
Objective: To investigate the effects of prostaglandin E2 (PGE2) and interleukin-1 (IL1-β) on the proliferation of cultured rabbit uterine ligament fibroblasts in vitro and in vivo. METHODS: The third generation of rabbit uterus ligament fibroblasts were treated with PGE2 (25, 50, 100, 500 and 1000ng / ml) and IL1-β (10,20,50,100,200 ng / ml) The combined stimulation group was experimental group, and the conventional culture group was used as the control group. The cell proliferation effect was determined by MTT assay. Results: When applied alone, 100 ng / ml PGE2 and 20 ng / ml IL1-β were the most effective proliferative concentrations (P <0.01), respectively. Compared with the control group (P <0.05). CONCLUSION: PGE2 and IL1-β in proper concentration can effectively promote the proliferation of uterine ligament fibroblasts in vitro, indicating that PGE2 and IL1-β can regulate the proliferation of uterine ligament fibroblasts.
