为了建立长白山地区槲寄生总生物碱的质量控制标准,试验以硫酸阿托品作为对照品建立了高效液相色谱法(HPLC),以十八烷基硅烷键合硅胶为填充剂(4.6 mm×250.0 mm,5μm)、0.05 mol/L的磷酸二氢钾溶液(含0.002 5 mol/L的辛烷磺酸钠)-乙腈(84∶16)为流动相,在流速为1.0 m L/min、波长为207 nm、柱温为30℃时,进样量设定为10μL,测定长白山槲寄生总生物碱含量。结果表明:槲寄生生物碱浓度与峰面积呈现良好的线性关系,其线性方程为y=31 527 561x-15 238,R2=0.999 4。说明HPLC法简便、准确、快速、专属性强、灵敏度高,可用于长白山地区槲寄生总生物碱含量的测定。
In order to establish the quality control standard of total alkaloids of mistletoe in Changbai Mountain, high performance liquid chromatography (HPLC) was established with atropine sulfate as the reference substance. The octadecylsilane bonded silica gel was used as the filler (4.6 mm × 250.0 mm , 5 μm), 0.05 mol / L potassium dihydrogen phosphate solution (containing 0.002 5 mol / L sodium octane sulfonate) -acetonitrile (84:16) as the mobile phase at a flow rate of 1.0 m L / 207 nm, the column temperature was 30 ℃, the injection volume was set to 10μL, determination of total Changbai Mountain mistletoe alkaloid content. The results showed that there was a good linear relationship between the alkaloid concentration and the peak area of the mistletoe. The linear equation was y = 31 527 561x-15 238, R2 = 0.999 4. The HPLC method is simple, accurate, rapid, specific and sensitive and can be used for the determination of total alkaloid content in Mistletoe of Changbai Mountain.
