目的:优选地龙的渗漉工艺参数。方法:采用体外溶血法,以含固量和蛋白酶活性为综合评价指标,通过单因素试验优选地龙的渗漉工艺。结果:尿激酶在25~125 U·mL-1与溶圈面积呈良好线性关系,最佳渗漉工艺参数为地龙粉碎为粗粉,浸泡时间0.5 h,渗漉速度2 mL·min-1,加8倍量60%乙醇渗漉;渗漉液体积220 mL,含固量3.252 g,蛋白酶活性5 624.7 U·g-1。结论:纤维蛋白原平板法可快速、简便、准确地测定地龙中酶的活性,优选的工艺参数稳定可行,为地龙有效成分的充分利用提供新选择。
Purpose: Optimum dragon percolation process parameters.
