目的 研究ER阳性和ER阴性的人乳腺癌细胞株整合素亚型α5β1、α4β1和组织蛋白酶D蛋白的表达和细胞生物学特性的关系。 方法 采用细胞培养、Southern杂交、Northern杂交和免疫组织化学技术,检测ER阳性MCF7及ER转染阳性MDAMB231和ER阴性MDAMB231人乳腺细胞中整合素亚型α5β1、α4β1和组织蛋白酶D蛋白的表达,了解它与细胞生物学特性的关系。 结果 MCF7、ER转染阳性MDAMB231和MDAMB231细胞中整合素亚型α5β1基因DNA,mRNA表达量各不相同。整合素亚型α5β1、α4β1和组织蛋白酶D蛋白在三株人乳腺癌细胞中的定位各不相同,且存在数量上的差别(P<005)。三苯氧胺具有诱导α5β1表达的作用,而对α4β1和组织蛋白酶D无影响。 结论 雌激素受体的表达和整合亚型α5β1、α4β1和组织蛋白酶D状态有关,三苯氧胺具有诱导转移相关基因表达的作用
Objective To investigate the relationship between the expression of integrin subtypes α5β1, α4β1 and cathepsin D and the biological characteristics of ER-positive and ER-negative human breast cancer cell lines. Methods Cell culture, Southern blot, Northern blot and immunohistochemical techniques were used to detect the expression of integrin subtypes α5β1, α4β1 and cathepsin D in ER positive MCF7 and ER transfected positive MDAMB231 and ER negative MDAMB231 human breast cells. It is related to the biological characteristics of cells. Results The mRNA and mRNA expression of integrin subtype α5β1 in MCF7 and ER transfected positive MDAMB231 and MDAMB231 cells were different. The localization of integrin subtypes α5β1, α4β1 and cathepsin D protein in three human breast cancer cells was different and there was a quantitative difference (P<005). Tamoxifen had the effect of inducing α5β1 expression but had no effect on α4β1 and cathepsin D. Conclusion The expression of estrogen receptor is related to the integration of subtypes α5β1, α4β1 and cathepsin D. Tamoxifen has the effect of inducing the expression of metastasis-associated genes.
