目的:探讨 BRCA_1蛋白在乳腺癌组织中的表达以及与乳腺癌临床病理的关系,了解 BR-CA_1蛋白的表达与 P53及 C-erbB_2表达之间的相关性。方法:采用 LDP 免疫组化法对60例乳腺癌,10例乳腺良性疾病患者的石腊标本进行了 BRCA_1蛋白、P53、C-erbB_2的联合检测,结合患者的年龄、家族史、病理组织学分级,腋淋巴结的转移情况,雌、孕激素受体状况等因素进行相关分析。结果:BRCA_1蛋白的阳性表达率在恶性组为61.66%(37/60),在良性组表达为0(0/10)。BRCA_1蛋白的表达绝大部分为浆表达。BRCA_1蛋白的表达与年龄呈负相关(r=-0.295,P<0.05),与家族史呈正相关(r=0.509,P<0.01),BRCA_1蛋白的表达在不同组织学分级中(Ⅰ-Ⅲ)存在差异(P<0.05),随着组织学分级的增加 BRCA_1蛋白阳性表达的程度也增高(r=0.413,P<0.01),BRCA_1蛋白的表达在有无腋淋巴结转移的病例中存在差异(P<0.05),随着腋淋巴结转移数目的增加,BRCA_1蛋白阳性表达的程度也增高(r=0.365,P<0.01)。BRCA_1蛋白的表达与 ER、PR 之间以及与 P53及 CerbB_2表达之间本研究未见其相关性。结论:BRCA_1蛋白的表达与年轻、有家族史,组织学分级高,腋淋巴结转移相关,随着进一步研究的深入,推测 BRCA_1蛋白的检测将作为估计乳腺癌生物学行为和预后的新指标。
OBJECTIVE: To investigate the expression of BRCA_1 protein in breast cancer tissues and the clinicopathological features of breast cancer, and to investigate the correlation between the expression of BR-CA_1 protein and the expression of P53 and C-erbB_2. Methods: The specimens of paraffin embedded in 60 cases of breast cancer and 10 cases of breast benign disease were examined by LDP immunohistochemistry. The combined detection of BRCA_1 protein, P53 and C-erbB_2 was performed. The patients’ age, family history and histopathological grade , Axillary lymph node metastasis, estrogen and progesterone receptor status and other factors related analysis. Results: The positive rate of BRCA_1 protein was 61.66% (37/60) in malignant group and 0 (0/10) in benign group. The majority of BRCA_1 protein expression is in plasma. The expression of BRCA_1 protein was negatively correlated with age (r = -0.295, P <0.05) and positively correlated with family history (r = 0.509, P <0.01). The expression of BRCA_1 protein in different histological grades (Ⅰ-Ⅲ) (P <0.05). The positive expression of BRCA_1 protein was also increased with the increase of histological grade (r = 0.413, P <0.01). There was a significant difference in the expression of BRCA_1 protein in cases with axillary lymph node metastasis <0.05). As the number of axillary lymph node metastasis increased, the positive expression of BRCA_1 protein increased (r = 0.365, P <0.01). No correlation was found between the expression of BRCA_1 protein and ER, PR and between P53 and CerbB_2. CONCLUSIONS: The expression of BRCA_1 protein is associated with young, family history, histological grade and axillary lymph node metastasis. With further research, the detection of BRCA_1 protein will be a new index for estimating the biological behavior and prognosis of breast cancer.
