目的探讨CD147与基质金属蛋白酶(metalloprotease,MMP)-9在乳腺浸润性导管癌组织中的表达及意义。方法应用免疫组织化学SP法检测60份乳腺浸润性导管癌石蜡包埋组织中CD147与MMP-9的表达;采用Western blot方法检测28份乳腺浸润性导管癌及癌旁新鲜组织标本中CD147与MMP-9表达情况。结果免疫组织化学结果显示,CD147主要在癌细胞的胞膜及胞质中表达,在间质中不表达;MMP-9主要在癌细胞的胞质中表达,在间质中不表达;CD147与MMP-9表达在组织学分级及淋巴结转移上差异有统计学意义(P<0.05);在年龄、肿块大小及临床分期上差异无统计学意义(P>0.05);2种蛋白在乳腺浸润性导管癌组织中表达有明显相关性(r=0.769,P<0.05)。Western blot结果显示,乳腺浸润性导管癌组织中CD147与MMP-9表达高于癌旁乳腺组织(P<0.05),且有淋巴结转移者高于无淋巴结转移者(P<0.05)。结论 CD147与MMP-9在乳腺浸润性导管癌组织中表达明显增高,可能在乳腺癌发生、发展中发挥重要作用。
Objective To investigate the expression and significance of CD147 and metalloprotease (MMP) -9 in breast invasive ductal carcinoma. Methods The expression of CD147 and MMP-9 in paraffin-embedded tissues of 60 infiltrating invasive ductal carcinomas was detected by immunohistochemical SP method. The expressions of CD147 and MMP-9 in tissues from 28 infiltrating ductal breast carcinomas and adjacent tissues were detected by Western blot -9 expression. Results The results of immunohistochemistry showed that CD147 was mainly expressed in the cytoplasm and cytoplasm of cancer cells but not in the stroma. MMP-9 was mainly expressed in the cytoplasm of cancer cells but not in interstitial cells. CD147 and The expression of MMP-9 in histological grade and lymph node metastasis had statistical significance (P <0.05), but no significant difference in age, tumor size and clinical stage (P> 0.05) The expression of VEGF in ductal carcinoma was significantly correlated (r = 0.769, P <0.05). The results of Western blot showed that the expressions of CD147 and MMP-9 in invasive ductal carcinoma of breast tissue were higher than that in adjacent non-cancerous tissues (P <0.05), and those with lymph node metastasis were higher than those without lymph node metastasis (P <0.05). Conclusion The expressions of CD147 and MMP-9 in invasive ductal carcinoma of the breast are significantly increased, which may play an important role in the occurrence and development of breast cancer.
