目的:观察人α-防御素HNP1基因对人鼻咽癌细胞株HNE-1生长的影响,探讨其在裸鼠鼻咽癌移植瘤生长中的作用。方法:HNP1基因通过脂质体介导转染人鼻咽癌HNE1细胞株,四甲基偶氮唑蓝(MTT)实验观察对HNE-1细胞的细胞毒性作用,将筛选的含pcDNA3.1(+)-HNP1质粒的HNE-1细胞和空pcDNA3.1(+)质粒的HNE-1细胞以及单纯的HNE-1细胞培养、收集后分别注射于6周龄雌性BALB/c裸鼠的右侧腹壁皮下,观察肿瘤生长速度,免疫组织化学检测α-防御素1在肿瘤组织中的表达。结果:HNP1基因转入鼻咽癌HNE-1细胞后能成功地表达有活性的α-防御素1,MTT实验证实有细胞毒作用,转染HNP1基因组裸鼠较对照组肿瘤生长速度慢,体积小。结论:通过转基因方法能表达有活性的α-防御素1并能抑制裸鼠鼻咽癌移植瘤的生长。
OBJECTIVE: To observe the effect of human α-defensin HNP1 gene on the growth of human nasopharyngeal carcinoma cell line HNE-1 and to explore its role in the growth of nasopharyngeal carcinoma in nude mice. Methods: HNP1 gene was transfected into human nasopharyngeal carcinoma HNE1 cell line by lipofectamine. The cytotoxicity of HNP1 gene on HNE-1 cells was observed by MTT assay. +) - HNE-1 cells of HNP1 plasmid and HNE-1 cells of empty pcDNA3.1 (+) plasmid and HNE-1 cells were cultured and collected and then injected into the right side of 6-week-old female BALB / c nude mice Subcutaneous abdominal wall, observe the growth rate of tumor, immunohistochemical detection of α-defensin 1 expression in tumor tissue. RESULTS: The HNP1 gene was successfully transfected into HNE-1 cells of nasopharyngeal carcinoma and successfully expressed the active α-defensin 1. MTT assay confirmed that the HNP1 gene transfected HNP1 gene in nude mice had slower tumor growth rate and volume small. Conclusion: The transgenic α-defensin 1 can be expressed and inhibited the growth of nasopharyngeal carcinoma in nude mice.
