准确的HLA配型是异基因造血干细胞移植 (Allo -HSCT)成功的关键之一 ,经典的血清学方法用于HLA -II类抗原的配型因存在诸多不足可导致漏配或错配 ,为了克服血清学方法的种种不足 ,我们建立了HLA -II类聚合酶链反应 -序列特异性引物 (PCR -SSP)基因配型方法 .此方法可避开血清学方法的种种干扰 ,直接从基因水平检测HLA的个体遗传学差异 .结果表明 :PCR -SSP方法具有重复性好、特异性强、灵敏度高、实验结果易于判断等特点 ,对于Allo -HSCT选择理想供者 ,减少或减轻Allo -HSCT时GVHD的发生 ,有着至关重要的意义 .可满足Allo -HSCT对HLA配型准确性高的要求 ,为在同胞兄弟姐妹间和无血缘关系供 -受者之间选择理想供者提供了重要保证
