目的 获得能早期诊断乙型肝炎病毒(HBV)相关性肝细胞癌(HCC)的癌组织蛋白质组模式。方法 应用双向凝胶电泳(2-DE)技术获得癌组织和非癌组织蛋白质表达谱。先对14例癌组织和非癌组织蛋白质表达谱进行分析,在癌组织蛋白质表达谱中发现具有鉴别癌组织和非癌组织的蛋白质组模式,再应用该模式对48例未知的癌组织和非癌组织2-DE图谱进行检测。结果 未知的癌组织与非癌组织均得到准确鉴定,灵敏度和特异性均为100%。结论 对蛋白质组模式中的蛋白质点进一步分析有助于研究HBV相关性HCC的肿瘤标记。蛋白质组模式技术亦有望成为在高危人群和普通人群中对恶性肿瘤进行筛查的工具。
Objective To obtain proteomic patterns of cancer tissues that can early diagnose hepatitis B virus (HBV) -related hepatocellular carcinoma (HCC). Methods Two-dimensional gel electrophoresis (2-DE) was used to obtain the protein expression profile of cancer and non-cancer tissues. First of all 14 cases of cancerous and non-cancerous tissue protein expression profiles were analyzed in the cancer tissue protein profile was identified with cancer tissue and non-cancerous tissue proteome pattern, and then applied to 48 cases of unknown cancer and non-cancerous tissue Cancer tissue 2-DE pattern was tested. The results of unknown cancer and non-cancerous tissue were accurately identified with both sensitivity and specificity of 100%. Conclusion Further analysis of protein spots in the proteomic model is useful for studying tumor markers of HBV-associated HCC. Proteomics is also expected to be a tool to screen for malignancies in high-risk and general-population populations.
