[目的 ]对河南新乡一起6 0 Co源辐射事故中 3例受照者进行了CB微核分析 ,并对其生物剂量进行估算。 3例受照者A、B、C均为男性 ,受照时年龄分别为 3 1、3 9和 51岁 ,分别受照后2 4d、44d和 96d采血培养 ,进行CB微核分析和剂量估算。 [方法 ]CB微核标本的制备方法为 :对 3例受照者分别取 0 3~ 0 4ml肝素抗凝静脉血 ,加到 4ml含 2 0 %小牛血清和 50 μg/mlPHA的混合培养液中 ,置 3 7℃恒温培养箱中培养 ,培养至 40~ 44h ,加入终浓度为 6mg/L的松胞素B ,继续培养至 72h收获。常规制片 ,Giemsa染色。显微镜下观察双核淋巴细胞 ,并记录其微核数。结果以微核细胞率 (CBCMN ,‰ )和微核率 (CBMN ,‰ )表示 ,并估算剂量。生物剂量估算采用白玉书教授等建立的6 0 Coγ射线照射离体人血诱发的CB微核的剂量曲线估算生物剂量。[结果 ]照后 2 4d 3例受照者的CB微核率和CB微核细胞率均明显升高 ,由此估算的剂量分别为 :① 3例受照者依据CBMN率估算的个体辐射剂量分别为 0 97Gy(A)、0 74Gy(B)和 0 58Gy(C) ;②依据CBCMN率估算的个体辐射剂量分别为 0 87Gy(A)、0 64Gy(B)和 0 50Gy(C)。 [结论 ]本文用CBMN估算剂量与染色体“双 +环”畸变剂量和物理剂量接近 ,与放射损伤的临床诊断亦相符合。说明CB微核分析估?
[Objective] To study the micronucleus test of CB in three co-radiation incidents involving 6 0 Co source in Xinxiang, Henan Province, and estimate their biological dose. A, B, and C were male, and the age at irradiation was 3 years, 13 years and 51 years respectively. Blood samples were collected at 24, 44 and 96 days after irradiation respectively. [Method] The preparation methods of CB micronucleus specimens were as follows: 0 3 ~ 0 4 ml heparin anticoagulation venous blood was taken from 3 irradiated subjects and added to 4 ml mixed medium containing 20% calf serum and 50 μg / ml PHA Medium and set at 37 ℃ incubator cultured to 40 ~ 44h, adding a final concentration of 6mg / L of cytochalasin B, continue to culture until 72h harvest. Conventional production, Giemsa staining. Under the microscope, binucleated lymphocytes were observed and their micronuclei were recorded. The results were expressed as micronuclei (CBCMN, ‰) and micronuclei (CBMN, ‰), and the dose was estimated. Biological dosimetry The biological dosimetry was estimated using dose curves of 60 micronucleated CB micro-nuclei irradiated by 60 Co γ-rays established by Professor Bai Yushu et al. [Results] The CB micronucleus rate and CB micronucleus rate of 3 irradiated subjects were significantly increased at 24 days after irradiation. The estimated doses were as follows: ① The individual dose of radiation (0 97Gy (A), 0 74Gy (B) and 0 58Gy (C) respectively); (2) The individual radiation doses estimated by CBCMN rate were 0 87Gy (A), 0 64Gy (B) and 0 50Gy (C) respectively. [Conclusion] In this paper, CBMN dose is estimated to be close to chromosome “double + ring” distortion dose and physical dose, which is also consistent with the clinical diagnosis of radiation injury.
