[目的]探讨奥曲肽作用前后人肺腺癌A-549细胞系生长抑素受体各亚型蛋白和mRNA的表达并分析其表达变化的临床意义以及奥曲肽对细胞株增殖的影响。[方法]免疫组化法测定人肺腺癌A-549细胞系中5种生长抑素受体亚型的表达;RT-PCR检测奥曲肽作用前后生长抑素受体亚型mRNA的表达,并进行半定量分析;MTT法检测奥曲肽对细胞株增殖的影响。[结果]人肺腺癌A-549细胞系中5种生长抑素受体亚型(SSTR1~SSTR5)均有表达,奥曲肽抑制人肺腺癌A-549细胞系增殖呈现浓度依赖性;人肺腺癌A-549细胞系表达5种生长抑素受体亚型的mRNA和蛋白,SSTR3和SSTR1的表达强度明显高于SSTR2、SSTR4、SSTR5(P<0.05)。奥曲肽作用后SSTR1的mRNA表达量明显高于其他4型,SSTR2、SSTR5mRNA的表达量明显高于对照组的mRNA表达量(P<0.05)。[结论]5种生长抑素受体亚型在人肺腺癌A-549细胞系中均有表达,八肽生长抑素奥曲肽可以明显抑制肺腺癌细胞系的增殖。SSTR1、SSTR2、SSTR5在抑制人肺腺癌A-549细胞系生长和增殖过程中发挥了关键性作用。
[Objective] To investigate the clinical significance of the expression of somatostatin receptor subtypes protein and mRNA in human lung adenocarcinoma A-549 cell line before and after octreotide administration and the effect of octreotide on cell proliferation. [Method] The expression of 5 somatostatin receptor subtypes in human lung adenocarcinoma A-549 cell line was detected by immunohistochemistry. The expression of somatostatin receptor subtypes mRNA was detected by RT-PCR before and after octreotide treatment Semi-quantitative analysis; MTT assay octreotide impact on cell proliferation. [Results] The five somatostatin receptor subtypes (SSTR1 ~ SSTR5) in human lung adenocarcinoma A-549 cell line were all expressed. Octreotide inhibited the proliferation of A-549 cell line in a concentration-dependent manner. The expression of SSTR3 and SSTR1 was significantly higher in adenocarcinoma A-549 cell line than in SSTR2, SSTR4 and SSTR5 (P <0.05). The mRNA expression of SSTR1 in octreotide was significantly higher than that in other 4 types. The mRNA expression of SSTR2 and SSTR5 in octreotide was significantly higher than that in control (P <0.05). [Conclusion] All five somatostatin receptor subtypes are expressed in human lung adenocarcinoma cell line A-549. Octreotide octotide inhibits the proliferation of lung adenocarcinoma cell line. SSTR1, SSTR2 and SSTR5 play key roles in inhibiting the growth and proliferation of human lung adenocarcinoma A-549 cell line.
