响应面分析南方红豆杉枝条多糖超声提取工艺及其抗氧化作用研究

目的:研究南方红豆杉枝条中多糖的超声提取工艺及抗氧化作用。方法:在单因素实验基础上,采用响应面分析法对影响南方红豆杉枝条中多糖提取率的主要因素进行优化,建立影响因素与响应值之间的数学模型,确定最佳超声提取工艺条件;通过DPPH自由基清除率、还原力和总的抗氧化活性的测定,分析其抗氧化作用。结果:最佳超声提取工艺为:料液比26.78(V/W),提取时间42.81min,超声频率为33.88kHz,提取2次;南方红豆杉多糖对DPPH自由基具有一定的清除作用,总的抗氧化活性和还原能力均随多糖浓度的增加而上升。结论:采用该方法对提取条件进行优化,合理可靠,能为生产工艺提供理论依据;其多糖具有较强的抗氧化作用。 Objective: To study the ultrasonic extraction process and antioxidation of polysaccharides in Taxus chinensis branches. Methods: Based on the single factor experiment, the main factors influencing the extraction rate of polysaccharide in Taxus chinensis var. Mairei branches were optimized by response surface methodology. The mathematical model between the influencing factors and the response value was established to determine the optimal ultrasonic extraction conditions. The DPPH radical scavenging activity, reducing power and total antioxidative activity were assayed for antioxidant activity. Results: The optimal ultrasonic extraction process was as follows: the ratio of material to liquid was 26.78 (V / W), the extraction time was 42.81min, the ultrasonic frequency was 33.88kHz, extracted twice. The polysaccharide from S. australis had certain scavenging effect on DPPH free radical, Antioxidant activity and reducing ability increased with the increase of polysaccharide concentration. Conclusion: The extraction conditions are optimized by this method, which is reasonable and reliable. It can provide a theoretical basis for the production process and its polysaccharide has a strong antioxidant effect.
