目的探讨生长抑素及其类似物对人肝癌细胞的抑制作用及其对周期素依赖性蛋白激酶5(CDK5)的表达的影响。方法体外培养人肝癌细胞株(HepG2),在倒置显微镜下观察不同浓度组肝癌细胞的形态变化。MTT比色实验观察不同浓度的奥曲肽(Octreotide)对细胞增殖的影响。细胞爬片,免疫细胞化学法检测CDK5的表达情况。结果不同浓度的奥曲肽对肝癌细胞增殖的抑制作用有差异,高剂量组的抑制作用最强。药物作用48h后,药物组CDK5的表达较对照组明显减少,着色程度随药物浓度增高而逐渐减弱。结论奥曲肽对肝癌细胞的增殖有抑制作用,可下调肝癌细胞中CDK5的表达,CDK5在奥曲肽抑制肝癌细胞增殖的过程中发挥作用。
Objective To investigate the inhibitory effect of somatostatin and its analogues on human hepatoma cells and its effect on the expression of cyclin dependent protein kinase 5 (CDK5). Methods Human hepatocellular carcinoma cell line HepG2 was cultured in vitro. The morphological changes of hepatocarcinoma cells in different concentrations were observed under an inverted microscope. MTT assay was used to observe the effect of octreotide with different concentrations on cell proliferation. CLC was used to detect the expression of CDK5 by immunocytochemistry. Results Different concentrations of octreotide had different inhibitory effects on the proliferation of hepatocarcinoma cells, while the inhibitory effect of the high dose group was the strongest. After 48h, the expression of CDK5 in the drug group was significantly reduced compared with that in the control group, and the degree of coloration decreased with the increase of the drug concentration. Conclusion Octreotide can inhibit the proliferation of hepatoma cells and down-regulate the expression of CDK5 in hepatoma cells. CDK5 plays a role in the inhibition of octreotide on the proliferation of hepatoma cells.
