目的研究阿魏酸对血管平滑机细胞增殖的影响。方法采用贴块法培养人脐动脉血管平滑肌细胞(VSMC);台盼兰染色法检测药物对细胞的毒性;以四甲基偶氮唑蓝(MTT)法观察药物对血清诱导的VSMC增殖及生长的影响;以Bradford法检测细胞总蛋白含量变化;用5溴-2脱氧尿苷(BrdU)免疫酶联吸附法观察对DNA合成的影响。结果阿魏酸可逆性的延迟细胞对数生长期,影响细胞生长;有效抑制细胞的增殖活性,当阿魏酸终浓度为100μmol/L时,对细胞增殖的抑制率达到61.32%;引起细胞总蛋白的合成及BrdU的DNA掺入量下降,影响细胞周期。结论阿魏酸有效抑制血清诱导的VSMCs增殖,对以VSMCs异常增殖为靶标的动脉粥样硬化等心脑血管疾病的预防和治疗具有积极作用。
Objective To study the effect of ferulic acid on the proliferation of vascular smooth muscle cells. Methods Human umbilical artery smooth muscle cells (VSMCs) were cultured by patch method. The cytotoxicity of the drugs on cells was detected by trypan blue staining. The proliferation and growth of VSMC induced by drugs were observed by methyl thiazolyl tetrazolium (MTT) The changes of total cellular protein were detected by Bradford method. The effect of DNA synthesis was observed by BrdU immunosorbent assay. Results The reversible ferulic acid delayed the cell growth in the logarithmic growth phase and inhibited the cell proliferation. When the final concentration of ferulic acid was 100 μmol / L, the cell proliferation rate was 61.32% Protein synthesis and BrdU DNA incorporation decreased, affecting the cell cycle. Conclusion Ferulic acid can effectively inhibit the proliferation of VSMCs induced by serum and play an active role in the prevention and treatment of cardiovascular and cerebrovascular diseases such as atherosclerosis which target abnormal proliferation of VSMCs.
