系统地研究了高锰酸钾对 6 巯基嘌呤的氧化作用 ,提出了高灵敏测定 6 巯基嘌呤的荧光光度分析新方法。在碱性介质中 ,氧化产物的荧光强度较 6 巯基嘌呤自身的荧光强度提高了 1 0倍 ,且稳定性好。本体系的激发波长为 2 86nm,发射波长为 397nm,荧光强度与 6 巯基嘌呤的浓度在 0 .0 0 64~ 3.0μg m L范围内呈线性关系 ,检测限为 0 .0 0 32μg m L。应用该法于片剂中 6 巯基嘌呤含量的测定 ,与标准方法对照 ,结果令人满意。
The oxidative effect of potassium permanganate on 6-mercaptopurine has been systematically studied. A new method for the determination of 6-mercaptopurine by fluorescence spectrophotometry has been proposed. In alkaline medium, the fluorescence intensity of the oxidation product is 10 times higher than the fluorescence intensity of 6-mercaptopurine itself, and the stability is good. The excitation wavelength of this system is 2 86nm and the emission wavelength is 397nm. The fluorescence intensity is linear with the concentration of 6-mercaptopurine in the range of 0.064 ~ 3.0μg m L with the detection limit of 0.00322μg m L. Application of the method in the determination of 6-mercaptopurine tablets, compared with the standard method, the results are satisfactory.
