采用水加热回流法制备木香供试品溶液,以乙腈-0.1%甲酸梯度洗脱,检测波长280 nm,建立了木香水提液的HPLC指纹图谱,并对18批不同产地和不同采收期的木香药材进行相似度、主成分和聚类分析,结果表明不同产地和不同采收期的木香药材水溶性成分的差异较小。从18批样品的指纹图谱中确定了21个共有峰,通过与对照品比对,确认其中的6个特征峰分别为5-乙氧基甲基糠醛、紫丁香苷、绿原酸、咖啡酸、异绿原酸A和异绿原酸B。以绿原酸峰为参照峰,18批样品指纹图谱中各共有峰相对保留时间的RSD均小于0.36%。
The sample solution was prepared by refluxing with water and the gradient was eluted with acetonitrile-0.1% formic acid. The detection wavelength was 280 nm. Taking the peak of chlorogenic acid as the reference peak, the RSDs of the relative retention time of each common peak in 18 batches of samples were less than 0.36%.
