高赖氨酸玉米的单粒鉴别技术不仅要求准确而且要求简便以适应作大量的单粒分析。根据考马斯亮兰 G 250可以定量地与蛋白质相结合的原理,将点有各籽粒的醇溶蛋白提取液的滤纸先用考马斯亮兰染色,经脱色后在滤纸上就可以显现出深浅程度不同的色斑。色斑深的醇溶蛋白含量高赖氨酸含量低;色斑浅的醇溶蛋白含量低而赖氨酸含量高。这样,就可以用肉眼直接挑选出那些高赖氨酸籽粒。
