目的研究经游泳池水传播的腺病毒毒株血清型及基因特征。方法对采用呼吸道病毒多重PCR法检测阳性的16份咽试子样品进行了腺病毒3型、7型及21型的特异性PCR检测,并对其中3份标本进行了腺病毒六联体(hexon)基因和纤毛(fiber)基因的全序列测定,将测定结果与Genbank数据库公开的核酸序列进行同源性比对,绘制进化树。结果 16份腺病毒阳性样品均为腺病毒7型;3份样品间hexon基因和fiber基因核酸序列完全一致,同源性为100%;同时,该3份标本的hexon基因和fiber基因核酸序列与引起我国陕西省婴儿严重急性下呼吸道感染暴发的腺病毒毒株的核酸序列完全一致,并未发现明显的基因变异情况。结论本起经游泳池水传播的腺病毒暴发是由腺病毒7型引起,hexon基因和fiber基因的核酸序列无变异。
Objective To study the serotypes and genetic characteristics of adenovirus strains transmitted by swimming pool water. Methods A total of 16 pharyngeal samples with positive respiratory multi-PCR were detected by specific PCR for adenoviruses type 3, type 7 and type 21, and 3 of them were detected by hexon ) Gene and the fiber gene were sequenced, and the results of the analysis were aligned with the published nucleotide sequences of GenBank to generate the phylogenetic tree. Results The 16 positive samples of adenovirus were all adenovirus type 7. The hexon gene and the fiber gene of the 3 samples were completely identical with 100% homology. At the same time, the hexon gene and the fiber gene of the 3 samples were identical to The nucleotide sequence of the adenovirus strain that caused the outbreak of severe acute respiratory tract infection in infants in Shaanxi Province of China was completely consistent and no obvious gene mutation was found. Conclusions The outbreak of adenovirus that has been transmitted by swimming pool water is caused by adenovirus type 7, and there is no variation in the nucleotide sequence of hexon and fiber genes.
