经生物学方法鉴定,小麦-中间偃麦草异附加系Z1高抗我国特有的大麦黄矮病毒GPV株系,用104个随机引物对Z1及其亲本中7902的基因组进行RAPD分析,发现其中的引物OPS-16能从Z1中扩增1个约1.7Kb的特异性片段。用引物OPS-16扩增Z1、中7902、中间偃麦草、中5,同样也从Z1、中间偃麦草、中5中能稳定地扩增此1.7Kb片段。对Z1的抗、感病单株的自交后代进行RAPD分析,发现在所测的抗病株的自交后代中能稳定地扩增此1.7Kb片段,而在感病株的自交后代中则不能扩增。
The results showed that wheat-Thinopyrum intermedium Z1 was highly resistant to the barley yellow dwarf virus GPV strain in our country. RAPD analysis of the genome of 7902 in Z1 and its parents was carried out with 104 random primers. The results showed that the primers OPS-16 can amplify a specific fragment of about 1.7 Kb from Z1. The 1.7kb fragment was amplified from Z1, middle 7902, middle thinopyrum, middle 5 with the primer OPS-16 and also from Z1, The RAPD analysis of the selfing offspring of the resistant and susceptible Z1 plants showed that the 1.7Kb fragment could be stably amplified in the selfing offspring of the tested resistant plants and in the selfing offspring of susceptible plants In the can not be amplified.
