目的:探讨下调DNA解旋酶BLM对膀胱癌细胞化疗药物敏感性的影响。方法:采用脂质体转染技术将BLM-siRNA转染入膀胱癌细胞株RT112、5637和J82中,利用RT-qPCR和Westernblot方法检测BLM的mRNA和蛋白表达情况,借助CCK-8法检测吡柔比星和顺铂对膀胱癌细胞的半数抑制率浓度及化疗药敏感性。结果:吡柔比星和顺铂作用于三种膀胱癌细胞株,随着药物浓度的增加和作用时间的延长,三种膀胱癌细胞株的增殖逐渐降低。与对照组比较,下调膀胱癌细胞内BLM后,BLM的mRNA和蛋白表达均明显降低,且在吡柔比星和顺铂作用下细胞增殖较对照组也明显降低,差异均有统计学意义(P<0.05)。结论:下调BLM可增加人膀胱癌细胞对化疗药物的敏感性。
Objective: To investigate the effect of DNA helicase BLM on chemosensitivity of bladder cancer cells. Methods: BLM-siRNA was transfected into bladder cancer cell lines RT112, 5637 and J82 by liposome transfection technique. The mRNA and protein expression of BLM were detected by RT-qPCR and Western blot. The expression of BLM-siRNA was detected by CCK-8 The ratio of half inhibitory concentration of radiobasic and cisplatin on bladder cancer cells and chemosensitivity. Results: Pirarubicin and cisplatin were applied to three kinds of bladder cancer cell lines. With the increase of drug concentration and the prolongation of action time, the proliferation of the three bladder cancer cell lines gradually decreased. Compared with the control group, the BLM mRNA and protein expression were significantly decreased after BLM in bladder cancer cells was reduced, and the cell proliferation was also significantly decreased in the control group with pirarubicin and cisplatin, the differences were statistically significant ( P <0.05). Conclusion: Down-regulation of BLM can increase the sensitivity of human bladder cancer cells to chemotherapeutic drugs.
