目的探讨锌对慢性酒精中毒肝损伤再生的影响。方法健康成年雄性C57BL6小鼠40只,随机分为对照组、慢性酒精中毒、酒精中毒+锌及对照+锌4组,持续喂养6个月,形成慢性酒精中毒模型性肝病。测定血清和肝组织中锌和铁含量以及谷丙转氨酶(ALT)活性,免疫组织化学检测增殖细胞核抗原(PCNA)阳性细胞,逆转录-聚合酶链反应(RT-PCR)方法测定肝细胞核4α因子(HNF-4α)mPNA的表达变化。结果慢性酒精中毒小鼠肝组织锌含量为(6.83±0.82)μmol/L,明显减少,低于对照组的(15.71±1.63)μmol/L(P<0.01);与对照组ALT活性[(26.2±1.4)U/L]比较,慢性酒精中毒组ALT活性[(60.5±16.8)U/L]升高(P<0.05);补锌进一步增加了PCNA阳性细胞的数目,HNF-4 mRNA水平因酒精而明显减弱。结论补充锌能够增强慢性酒精中毒小鼠肝再生,其机制与肝细胞核4α因子(HNF-4α)的增加有关
Objective To investigate the effect of zinc on the regeneration of liver injury induced by chronic alcoholism. Methods Forty healthy adult male C57BL6 mice were randomly divided into control group, chronic alcoholism, alcoholism + zinc and control + zinc group, and continued to feed for 6 months to form model liver disease with chronic alcoholism. The contents of zinc and iron and the activity of alanine aminotransferase (ALT) in serum and liver tissue were determined. The proliferating cell nuclear antigen (PCNA) positive cells were detected by immunohistochemistry. The expressions of nuclear factor 4α (HNF-4α) mPNA expression changes. Results Compared with the control group, the zinc content in the liver of chronic alcoholism mice was (6.83 ± 0.82) μmol / L, which was significantly lower than that of the control group (15.71 ± 1.63) μmol / L (P <0.01) (60.5 ± 16.8) U / L] (P <0.05); zinc supplementation increased the number of PCNA positive cells, the level of HNF-4 mRNA Alcohol was significantly weakened. CONCLUSION: Zinc supplementation can enhance liver regeneration in mice with chronic alcoholism and its mechanism is related to the increase of hepatocyte nuclear factor 4α (HNF-4α)
