应用Dot-ELISA,以抗卫氏并殖吸虫囊蚴和成虫McAbG_2B_3、A_3D_3检测了4只Pw囊蚴感染犬吡喹酮治疗前后血清CAg动态。感染后第4d,4只犬囊蚴CAg均为阳性,1只犬成虫CAg为阳性。至第19d,4只犬囊蚴和成虫CAg均为阳性。第101～105d,4只犬中2只以每只3.2g总量吡喹酮进行治疗,治疗后短期内囊蚴和成虫CAg滴度明显升高,其中尤以成虫CAg升高显著。至治疗后第6d,囊蚴和成虫CAg达高峰,其后便逐步下降。1号犬在治疗后55d、2号犬于治疗后26d囊蚴CAg滴度为0.1号犬于治疗后65d、2号犬于治疗后36d成虫CAg滴度为0。未治疗犬成虫CAg维持于1:16～1:640,囊蚴CAg为1:20～1:40,CAg的动态呈现明显的期特异性。治疗后80d剖检2只治疗犬,其肺部均有多个陈旧性虫囊,虫囊内多为变性坏死虫卵,未发现成虫。未治疗犬于死后即行解剖检获Pw成虫60条。上述研究结果提示:1.感染Pw犬囊蚴CAg的动态具有明显的期特异性,与虫体在宿主体内的发育情况是一致的;2.有效治疗后囊蚴和成虫尤其是成虫CAg的滴度在短期内明显升高;3.可联合或单独应用抗囊蚴McAbG_2B_3、抗成虫McAbA_3D_3进行并殖吸虫病的虫期诊断和疗效考核。
Dot-ELISA was used to detect the dynamic changes of serum CAg before and after treatment with Praziquantel in 4 Pw larvae with McAbG_2B_3 and A_3D_3. On the 4th day after infection, CAg of 4 dogs were positive and CAg of 1 dog was positive. By the 19th day, four dogs, metacercaria and adult CAg were positive. From 101st to 105d, 2 of 4 dogs were treated with praziquantel (3.2g total), and the titers of metacercariae and adult CAg increased significantly after treatment, especially in adult CAg. At the 6th day after treatment, the metacercaria and adult CAg peaked, and then gradually decreased. The number of CAg in the No. 1 dog was 55 days after treatment, while that of the No. 2 dog in the No. 26 dog was 26 days after the treatment. The titer of the CAg in the No. 2 dog was 65 days after the treatment. The CAg of untreated dogs was maintained at 1: 16-1: 640 and the CAg of metacercariae was 1:20 to 1:40. The dynamic of CAg showed obvious period specificity. Eighty days after treatment, two dogs were dissected and two old obturatocysts were found in their lungs. Most of them were degenerative and necrotic eggs, and no adults were found. Untreated dogs were sacrificed 60 post-mortem Pw adult seized. The above results suggest that: 1. The dynamic period of CAg infection in Pw is obviously period-specific, which is consistent with the development of the host in vivo. 2. Effective treatment of post-metacercariae and adult worms CAg Degree in the short term was significantly increased; 3 can be combined or separate application of metacercariae McAbG_2B_3, anti-adult McAbA_3D_3 paragonimiasis pest diagnosis and efficacy assessment.
