目的:研究南海海洋真菌Penicillium sp.FS60的次级代谢产物及其细胞毒活性。方法:采用正相硅胶柱、反相硅胶柱、凝胶柱、HPLC和薄层制备等色谱技术和重结晶对菌株FS60的发酵产物进行分离纯化,并通过波谱分析进行结构鉴定;以神经胶质瘤细胞SF-268、乳腺癌细胞MCF-7、大细胞肺癌细胞NCI-H460为供试细胞株,采用SRB法测试化合物的细胞毒活性,以金黄色葡萄球菌(SA)、大肠杆菌(EC)和铜绿假单胞菌(PA)为指示菌株,采用改良的MMT法测试化合物的抗菌活性。结果:从发酵物中分离并鉴定了7个化合物,分别为:2,4-二羟基-3,5,6三甲基苯甲酸甲酯(1)、对羟基苯乙酮(2)、5-羟甲基糠酸(3)、isochromophilone VIII(4)、麦角甾醇(5)、过氧化麦角甾醇(6)、啤酒甾醇(7)。结论:化合物1为首次从青霉属真菌中分离得到,化合物3具有较强的抗菌活性,化合物4具有显著的胞毒活性。
Objective: To study the secondary metabolites of Penicillium sp. FS60 and its cytotoxic activity in the South China Sea. Methods: The fermentation products of strain FS60 were isolated and purified by normal phase silica gel column, reverse phase silica gel column, gel column, HPLC, TLC and recrystallization. The structures were identified by spectroscopic analysis. The cytotoxic activity of the compounds was tested by SRB method. The inhibitory effects of the compounds on the cytotoxicity of Staphylococcus aureus (SA), Escherichia coli (EC) And Pseudomonas aeruginosa (PA) as the indicator strains, the antibacterial activity of the compounds was tested by a modified MMT method. Results: Seven compounds were isolated and identified from the fermentation products as follows: 2,4-dihydroxy-3,5,6-trimethylbenzoate (1), p-hydroxyacetophenone (3), isochromophilone VIII (4), ergosterol (5), ergosterol peroxide (6) and beer sterol (7). Conclusion: Compound 1 is isolated from Penicillium fungi for the first time. Compound 3 has strong antibacterial activity and compound 4 has significant cytotoxic activity.
