目的探讨抗氧化物维生素E(Vitamin E)对人类体外培养胚胎凋亡及体外发育的影响,并观察培养过程中胚胎凋亡的形态学变化。方法以常规体外受精-胚胎移植(in vitro fertilization and embryotransfer,IVF-ET)后废弃的胚胎为材料,培养过程中添加抗氧化物质维生素E,观察其对胚胎体外发育的影响,采用TUNEL方法对胚胎进行染色,观察其凋亡情况;用Hoechst33342对胚泡进行荧光染色,根据胚泡细胞总数及凋亡形态学变化对其凋亡进行检测。结果当添加100μmol维生素E时,与对照组相比,显著提高人类扩张期囊胚形成率和孵化率(P<0.05),降低其凋亡率(P<0.05)。结论维生素E能够降低人类囊胚的凋亡率,提高其体外发育率。
Objective To investigate the effect of antioxidant vitamin E on apoptosis and in vitro development of human embryos cultured in vitro and to observe the morphological changes of embryonic apoptosis in culture. Methods The embryos after in vitro fertilization and embryotransfer (IVF-ET) were used as materials. Antioxidant vitamin E was added during the culture to observe its effect on embryo development in vitro. TUNEL method was used to detect embryos The apoptosis of blastocyst was observed by Hoechst33342 staining, and the apoptosis of blastocyst was detected by the total number of blastocysts and morphological changes of apoptosis. Results When 100 μmol vitamin E was added, the rate of blastocyst formation and hatching rate (P <0.05) and the apoptosis rate (P <0.05) were significantly increased in human compared with the control group. Conclusion Vitamin E can reduce the apoptosis rate of human blastocysts and increase their in vitro development rate.
