目的研究增加Y-STR位点和HLA基因座的检测后,对二联体亲子鉴定亲子关系相对机会(RCP)值的影响。方法对二联体亲子鉴定案例除常规进行ABI AmpFLSTR IdentifilerTM试剂盒中的15个常染色体STR位点检测以外,增加ABI YfilerTM系统中的17个Y-STR或HLA-A、-B、-DRB1基因座的检测,并计算增加检测位点前后的亲子关系指数(PI)和RCP。结果经ABI IdentifilerTM试剂盒检测的135例二联体亲子鉴定,RCP值>99.99%案例为109例,占80.74%,其余的26例(19.26%)RCP值均>99.73%、但<99.99%,在这26例单亲案例中,18例“母-子”单亲案例增加HLA-A、-B、-DRB1基因座检测后,其中的15例RCP值>99.99%;8例“父-子”单亲案例增加了17个Y-STR检测后,RCP值则均超过了99.99%。结论增加Y-STR或HLA-A、-B、-DRB1基因座等遗传标记物的检测,可有效提高二联体亲子鉴定的RCP值,从而提高鉴定的准确性。
Objective To investigate the effect of paternity testing on the relative opportunity (RCP) value of paternity testing of paired daughters after increasing detection of Y-STR loci and HLA loci. Methods In addition to the 15 autosomal STR loci in the ABI AmpFLSTR IdentifilerTM kit routinely performed in the case of duplex paternity testing, 17 Y-STRs or HLA-A, -B, -DRB1 genes in the ABI YfilerTM system were added Seat test, and calculate the increase in test sites before and after the parent-child relationship index (PI) and RCP. Results 135 cases of duplex paternity test by ABI IdentifilerTM kit showed 109 RCP values> 99.99%, accounting for 80.74%. The remaining 26 cases (19.26%) had RCP values> 99.73% but <99.99% Among the 26 single-parent cases, 15 cases had an RCP value of> 99.99% after 18 cases of “mother-child” single-parent cases were detected with HLA-A, -B and -DRB1 loci. After 17 Y-STR tests were added to the single-parent case, the RCP values exceeded 99.99%. Conclusions Increasing the genetic markers such as Y-STR or HLA-A, -B, -DRB1 loci can effectively improve the RCP value of the paternity test and improve the accuracy of identification.
