用改进的斑点法检测柑桔黄龙病类细菌、香蕉束顶病毒(BBTV),均获满意结果.阳性反应在硝基纤维转移膜上形成深浅不等的褐色斑点.检测健康柑桔、香蕉粗汁液,获阴性结果,在上述固相载体上不形成有色物质.在缓冲液TBS中加入2%TritonX-100和1%国产蛋白片,作为改进后的封闭缓冲液,封闭硝基纤维转移膜上未结合抗原部位,效果较好.斑点法检测活体香蕉束顶病样品及-30℃冷冻保存6个月香蕉束顶病样品,同样获满意结果.斑点法快速、准确、简单、灵敏度高、特异性强,可用于植物检疫,存在于硝基纤维转移膜上的检疫结果可长期保存.
