目的探讨anoctamin 1(ANO1)表达对非小细胞肺癌(NSCLC)病理分期和复发的影响。方法Ⅰ-Ⅲ期NSCLC患者64例。采用qRT-PCR、Western blot和免疫组化检测肺癌组织和癌旁组织ANO1和表皮生长因子受体(EGFR)的表达;行肿瘤病理分级评定,随访术后的复发。结果 NSCLC存在ANO1的过表达。ANO1阳性组28例中,26例(93%)的EGFR表达阳性;ANO1阳性组的病理Ⅲ期病例的比例高于ANO1阴性组(50%vs.17%)(P<0.01)。随访1年,ANO1阳性组的复发率高于ANO1阴性组(39%vs.17%)(P<0.01)。结论 ANO1在NSCLC组织存在过表达。ANO1的过表达促进EGFR的过表达,提高NSCLC的病理分级评分和复发率。
Objective To investigate the effect of anoctamin 1 (ANO1) expression on the pathological stage and recurrence of non-small cell lung cancer (NSCLC). Methods Ⅰ-Ⅲ NSCLC patients 64 cases. The expressions of ANO1 and epidermal growth factor receptor (EGFR) were detected by qRT-PCR, Western blot and immunohistochemistry. Tumor pathological grading was used to assess the recurrence after surgery. Results There was overexpression of ANO1 in NSCLC. Among the 28 cases of ANO1 positive group, EGFR expression was positive in 26 cases (93%). The proportion of pathological stage Ⅲ in ANO1 positive group was higher than that in ANO1 negative group (50% vs 17%) (P <0.01). The follow-up of 1 year showed that the recurrence rate of ANO1 positive group was higher than that of ANO1 negative group (39% vs 17%) (P <0.01). Conclusion ANO1 is overexpressed in NSCLC tissues. Overexpression of ANO1 promotes EGFR overexpression and improves pathological grade score and recurrence rate of NSCLC.
