目的研究Calpain-10基因44位点单核苷酸多态性与海南黎族2型糖尿病(T2DM)胰岛素抵抗的关系,探讨Calpain-10与T2DM发病易感性之间的关系。方法选择海南黎族健康查体者54例为正常对照组,T2DM患者46例为T2DM组。测量所有研究对象的身高、体重、腰围、臀围,计算体重指数(BMI)、腰臀比(WHR)。并且空腹抽血测定胰岛素(Fins)、空腹血糖(FPG)。用稳态模型方法,计算胰岛素抵抗指数(IRI)及胰岛β细胞功能指数(HBCI)。对所有研究对象Calpain-10基因UCSNP-44多态性进行基因分型并计算其优势等位基因频率。结果①黎族人正常对照组UCSNP-44 TT基因型占78%,TC型占20%,CC型占2%;T等位基因频率为88%,C等位基因为12%。黎族人T2DM组UCSNP-44 TT基因型占76%,TC型占22%,CC型占2%;T等位基因频率为88%,C等位基因为12%。2型糖尿病组与正常对照组比较,UCSNP-44位点等位基因及基因频率差异无统计学意义(P>0.05)。按BMI≥25 kg/m2、BMI<25kg/m2分别比较,各组之间也无统计学差异(P>0.05)。②T2DM组内UCSNP-44 TT组与TC+CC组BMI及WHR、FPG、FIns、IR、HBCI均无明显差异(P>0.05)。结论本研究未发现Calpain-10基因UCSNP-44位点变异与T2DM及胰岛素抵抗发病相关,提示Calpain-10基因UCSNP-44并非海南黎族人群中T2DM常见相关发病基因。
Objective To investigate the relationship between the single nucleotide polymorphism (SNP) 44 of Calpain-10 gene and insulin resistance in type 2 diabetes mellitus (T2DM) of Hainan Li ethnicity and explore the relationship between Calpain-10 and susceptibility to T2DM. Methods 54 cases of Hainan Li nationality physical examination for the control group, 46 cases of T2DM patients with T2DM. Body height, weight, waist circumference, hip circumference, body mass index (BMI), waist-hip ratio (WHR) were measured for all subjects. Fasting blood was collected to measure insulin (Fins) and fasting blood glucose (FPG). Insulin resistance index (IRI) and pancreatic β-cell function index (HBCI) were calculated using the steady-state model method. All subjects were genotyped for the UCSNP-44 polymorphism of the Calpain-10 gene and their predominant allele frequencies were calculated. Results ①The TT genotype of UCSNP-44 accounted for 78% in the normal control group of Li nationality, TC type accounted for 20%, CC type accounted for 2%; T allele frequency was 88% and C allele was 12%. In Lebanon T2DM group, UCSNP-44 TT genotype accounted for 76%, TC genotype 22%, CC genotype 2%, T allele 88% and C allele 12%. There was no significant difference in allele and gene frequency between UCSNP-44 locus and type 2 diabetes mellitus (P> 0.05). According to BMI≥25 kg / m2 and BMI <25 kg / m2 respectively, there was no significant difference between the groups (P> 0.05). ② There was no significant difference in BMI, WHR, FPG, FIns, IR and HBCI between UCSNP-44 TT group and TC + CC group in T2DM group (P> 0.05). Conclusion The UCSNP-44 locus mutation of Calpain-10 gene is not associated with the pathogenesis of T2DM and insulin resistance in this study, suggesting that UCSNP-Calpain-10 gene is not a common T2DM gene in Hainan Li population.
