目的:探讨肝癌组织中泛素连接酶Cullin 4A(CUL4A)的表达情况及CUL4A对肝癌细胞生长的影响。方法:选择2013年1月—2015年12月72例肝癌组织标本及其癌旁组织标本和72例正常肝组织标本作为对照,免疫组化法检测肝癌组织、癌旁组织和正常肝组织中CUL4A的表达情况,用CUL4A-siRNA、NC-siRNA转染Hep-G2、QSG-7701肝癌细胞,CCK法检测细胞的增殖情况,PI染色、流式细胞术检测细胞周期,Transwell法检测细胞的侵袭迁移能力。结果:肝癌组织中CUL4A的表达明显高于癌旁组织和正常肝组织(t=48.657,P=0.000)。肝癌组织中CUL4A的表达和肝癌的分化程度、肿瘤直径、淋巴结转移、TNM分级有关(t=7.254、5.254、5.835、8.212,P=0.000、0.007、0.004、0.000)。转染CUL4A-siRNA的Hep-G2、QSG-7701肝癌细胞株在3 d和7 d时的OD值明显低于NC-siRNA组(t=3.132、5.264;3.152、5.834,P=0.037、0.008;0.041、0.011)。转染CUL4A-siRNA的Hep-G2、QSG-7701肝癌细胞株S期细胞比例低于NC-siRNA组(F=4.124、4.142;P=0.014、0.009)。转染CUL4A-siRNA的Hep-G2、QSG-7701肝癌细胞株侵袭转移能力低于NC-siRNA组(t=3.756、4.635;4.104、4.967,P=0.003、0.000;0.003、0.000)。结论:肝癌组织中CUL4A的表达升高,肝癌组织中CUL4A的表达与肝癌的分化程度、肿瘤直径、淋巴结转移、TNM分级有关,CUL4A能够促进肝癌细胞的增殖、细胞周期进程和侵袭迁移能力。
Objective: To investigate the expression of Cullin 4A (CUL4A) and the effect of CUL4A on the growth of hepatocellular carcinoma cells in hepatocellular carcinoma. The percentage of S phase cells in Hep-G2 and QSG-7701 hepatoma cell lines transfected with CUL4A-siRNA was lower than that in NC-siRNA group (F = 4.124, 4.142; P = 0.014,0.009). The ability of invasion and metastasis of Hep-G2 and QSG-7701 hepatocellular carcinoma cell lines transfected with CUL4A-siRNA was lower than that of NC-siRNA group (t = 3.756,4.635; 4.104,4.967, P = 0.003,0.000; 0.003,0.000). Conclusion: The expression of CUL4A in hepatocellular carcinoma tissue is increased. The expression of CUL4A in hepatocellular carcinoma is related to the differentiation, tumor diameter, lymph node metastasis and TNM grade in hepatocellular carcinoma. CUL4A can promote the proliferation, cell cycle progression and invasion and migration of hepatoma cells.
