目的:探讨内皮细胞(endothelialcells,ECs)对血管平滑肌细胞(vascularsmoothmusclecells,VSMCs)整合素β1(β1-integrin)表达的影响,为研究血管壁细胞功能提供理论依据。方法:应用血管ECsVSMCs联合培养系统,以WesternBlot和细胞免疫荧光技术,检测与ECs联合培养12h后VSMCs的β1-integrin表达和分布变化。结果:与ECs的联合培养,能明显增强VSMCs的β1-integrin蛋白表达,同时诱导VSMCs表面β1-integrin的激活。结论:ECs可能通过调节VSMCs的β1-integrin表达,从而影响VSMCs的功能。
Objective: To investigate the effect of endothelial cells (ECs) on the expression of integrin β1 (β1-integrin) in vascular smooth muscle cells (VSMCs), and to provide a theoretical basis for studying the function of vascular wall cells. Methods: The vascular endothelial growth factor (EC) -VSMCs co-culture system was used to detect the expression and distribution of β1-integrin in VSMCs by Western Blot and immunofluorescence technique. Results: Co-culture with ECs could significantly increase the expression of β1-integrin protein and the activation of β1-integrin on VSMCs. Conclusion: ECs may affect the function of VSMCs by regulating the expression of β1-integrin in VSMCs.
