目的 :探讨GIRK1在颞叶癫大鼠海马齿状回的表达及其意义。方法 :112只雄性SD大鼠随机分为实验组 (n =70 )与对照组(n =42 ) ,同时建立海人藻酸 (KA)颞叶癫模型。选取KA腹腔注射后 3、6、12、2 4、48h ,7、3 0d为研究的时间点。用原位杂交法及免疫组织化学法检测海马齿状回GIRK1mRNA及蛋白的表达。结果 :实验组大鼠海马齿状回GIRK1mRNA表达在致后 6h较对照组减少 ,而在致后至 7~ 3 0d较对照组增高。结论 :在颞叶癫的不同时期海马齿状回GIRK1表达的变化反映出颞叶癫的复杂性。
Objective: To investigate the expression of GIRK1 in hippocampal dentate gyrus of temporal lobe epilepsy rats and its significance. Methods: One hundred and twelve male Sprague-Dawley rats were randomly divided into experimental group (n = 70) and control group (n = 42). At the same time, kainic acid (KA) temporal lobe epilepsy model was established. Select KA intraperitoneal injection 3,6,12,2 4,48 h, 7,3 0 d after the study point in time. In situ hybridization and immunohistochemistry were used to detect the mRNA and protein expression of GIRK1 in dentate gyrus. Results: The expression of GIRK1mRNA in hippocampal dentate gyrus in experimental group decreased at 6h after reperfusion, and increased at 7 ~ 30d after reperfusion. CONCLUSION: The changes of GIRK1 expression in dentate gyrus at different stages of temporal lobe epilepsy reflect the complexity of temporal lobe epilepsy.
