目的 研究 5 -氟尿嘧啶 (5 - fluorouracil,5 - FU)体外诱导胃癌 SGC- 790 1细胞凋亡作用 ,及细胞凋亡与 Ki- 6 7抗原、核增殖抗原 (PCNA)、P5 3蛋白、Bax蛋白、死亡受体 Fas表达的关系。探讨 5 - FU诱导胃癌细胞凋亡作用的机制。方法 应用流式细胞仪和 3′- OH末端标记法 (TUNEL)检测凋亡细胞 ,电镜观察凋亡细胞的形态改变 ,免疫组织化学法检测细胞 Ki- 6 7、PC-NA、P5 3、Bax和 Fas表达。结果 SGC- 790 1细胞经 5 - FU诱导后 ,凋亡指数 (DI)明显增加 ,且具有剂量、时间依赖性。实验组细胞 Ki- 6 7、PCNA表达明显低于对照组 ,伴 Bax表达 ,而 P5 3、Fas则在诱导前后均无表达。结论 5 - FU具有诱导胃癌 SGC- 790 1细胞凋亡及抑制细胞增殖作用。 5 - FU诱导 SGC- 790 1细胞可能是通过 P5 3非依赖途径
Objective To study the apoptosis of gastric cancer SGC7901 cells induced by 5-fluorouracil (5-FU) in vitro, and apoptosis and Ki-67 antigen, nuclear proliferating antigen (PCNA), P53 protein, Bax protein. Relationship between death receptor Fas expression. To investigate the mechanism of 5-FU-induced apoptosis in gastric cancer cells. Methods Apoptotic cells were detected by flow cytometry and 3’-OH labeling (TUNEL). Morphological changes of apoptotic cells were observed by electron microscope. Ki-67, PC-NA, P5 3, Bax were detected by immunohistochemistry. And Fas expression. Results After induction of 5-FU in SGC-790 cells, the apoptotic index (DI) increased significantly in a dose- and time-dependent manner. The expression of Ki-67 and PCNA in the experimental group was significantly lower than that in the control group with Bax expression, while P53 and Fas expression were not observed before and after induction. Conclusion 5-Fu can induce gastric cancer SGC-790 1 cell apoptosis and inhibit cell proliferation. 5-FU-induced SGC-790-1 cells may be through the P53-independent pathway
