目的:研究丹酚酸A(SA-A)抗肝损伤、肝纤维化作用及其对Ⅰ型胶原基因表达的影响。方法:采用CCl_4诱导大鼠肝损伤及肝纤维化,期间予SA-A灌胃治疗,另设秋水仙碱组、丹参组作对照,6w后进行肝组织病理学和Ⅰ型胶原免疫组化观察,肝组织羟脯氨酸(Hyp)、血清ALT、AST和白蛋白含量测定,并以RT-PCR法检测肝组织Ⅰ型胶原mRNA。结果:SA-A可降低血清ALT、AST水平及肝组织Hyp含量,减轻肝纤维化程度与秋水仙碱、丹参一致。此外,SA-A组肝组织Ⅰ型胶原mRNA表达明显轻于模型组。结论:丹酚酸A有显著的抗肝损伤、肝纤维化作用,对损伤肝组织Ⅰ型胶原mRNA表达有明显抑制作用。
Objective: To study the effect of salvianolic acid A (SA-A) on hepatic injury and liver fibrosis and its effect on the expression of type I collagen gene. METHODS: Liver injury and hepatic fibrosis were induced by CCl_4 in rats. SA-A was given intragastrically during the treatment period. Colchicine group and Salvia miltiorrhiza group were used as controls. Liver histopathology and type I collagen immunohistochemistry were performed after 6 weeks. The liver tissue hydroxyproline (Hyp), serum ALT, AST and albumin content were measured, and liver type I collagen mRNA was detected by RT-PCR. Results: SA-A can reduce serum ALT, AST levels and liver tissue Hyp content, reduce liver fibrosis consistent with colchicine and Salvia miltiorrhiza. In addition, the mRNA expression of type I collagen was significantly lighter in the SA-A group than in the model group. Conclusion: Salvianolic acid A has significant anti-hepatotoxicity and hepatic fibrosis effects, and inhibits the expression of type I collagen mRNA in liver tissue.
