目的观察白花九里明提取液对小鼠血压、心率以及血清一氧化氮(NO)、内皮素-1(ET-1)浓度的影响,初步分析白花九里明对心血管系统影响及其机制。方法取50只小鼠,随机均分为5组,正常对照组给予0.85%Na Cl注射液,美托洛尔组给予0.3 mg/m L美托洛尔溶液,替米沙坦组给予0.5 mg/m L替米沙坦溶液,白花九里明低浓度组和高浓度组分别给予150 mg/m L和300 mg/m L白花九里明提取液,灌胃每天1次,共14 d。末次灌胃20 min测量小鼠血压和心率后,摘除眼球取血,检测血清NO、ET-1浓度。结果白花九里明低浓度组的收缩压、舒张压、心率和血清NO、ET-1浓度与正常对照组比较差异无统计学意义(P>0.05)。白花九里明高浓度组的收缩压和舒张压均明显低于正常对照组和白花九里明低浓度组(P<0.05,P<0.01),但与替米沙坦组差异无统计学意义(P>0.05);白花九里明低浓度组、替米沙坦组的心率与正常对照组差异无统计学意义(P>0.05),美托洛尔组的心率明显低于正常对照组(P<0.01),白花九里明高浓度组的心率明显低于替米沙坦组(P<0.01)而与正常对照组和美托洛尔组差异无统计学意义(P>0.05)。血清NO浓度:仅有白花九里明高浓度组明显高于正常对照组(P<0.05),其余组间比较差异无统计学意义(P>0.05);血清ET-1浓度:仅有白花九里明高浓度组和替米沙坦组明显低于正常对照组(P<0.05,P<0.01),其余组间比较差异无统计学意义(P>0.05)。结论白花九里明达到一定用量能降低动脉血压,但对心率没有明显影响,其降压作用可通过减少心搏出量和降低外周阻力来实现。
OBJECTIVE To observe the effects of Baihua Jiu-LING extract on blood pressure, heart rate, serum nitric oxide (NO) and endothelin-1 (ET-1) concentrations in mice and preliminary analysis of the effects of Baihua Jiuli on cardiovascular system and its mechanism. Methods Fifty mice were randomly divided into five groups. The normal control group was given 0.85% Na Cl injection, the metoprolol group was given 0.3 mg / m L metoprolol solution, and the telmisartan group was given 0.5 mg / m L telmisartan solution, low concentration of white Jiu Li Ming and high concentration group were given 150 mg / m L and 300 mg / m L white Jiu Li Ming extract, gavage once daily for 14 d. At the end of the last 20 min, the blood pressure and heart rate of the mice were measured, and the eyeballs were removed. The serum concentrations of NO and ET-1 were measured. Results There was no significant difference in systolic blood pressure, diastolic blood pressure, heart rate and serum NO and ET-1 concentrations between the low-dose group and the control group (P> 0.05). Systolic blood pressure and diastolic blood pressure were significantly lower than those in the control group and the control group (P <0.05, P <0.01), but there was no significant difference between the control group and the telmisartan group (P > 0.05). There was no significant difference in heart rate between telmisartan group and control group (P> 0.05). The heart rate of metoprolol group was significantly lower than that of normal control group (P <0.01) ). The heart rate was significantly lower in the Jiu Li Ming high concentration group than in the telmisartan group (P <0.01), but not significantly different from the normal control group and the metoprolol group (P> 0.05). The concentration of NO in serum was significantly higher than that in normal control group (P <0.05), but there was no significant difference among the other groups (P> 0.05). Serum ET-1 concentration was only in Baihuajiuli High concentration group and telmisartan group were significantly lower than the normal control group (P <0.05, P <0.01), the remaining group was no significant difference (P> 0.05). Conclusions Baghu Jiuli can reduce arterial blood pressure to a certain level, but has no obvious effect on heart rate. Its antihypertensive effect can be achieved by reducing cardiac output and decreasing peripheral resistance.
