目的检测慢性乙型肝炎患者外周血中HBV特异性CTL细胞表面杀伤细胞抑制性受体(KIR)的表达情况。方法利用MHC-Ⅰ-肽-五聚体(pentamer)技术结合流式三色分析技术,直接离体情况下检测(directex vivo)慢性乙型肝炎患者不同HBV特异性CTL细胞表面KIR的表达情况。结果在慢性乙型肝炎患者,KIR阳性的CD8+T细胞明显增加;KIR阳性的HBcAg(18-27)特异性CTL、HBeAg(335-343)特异性CTL、HBp(575-583)特异性CTL的百分比分别为12%、20%、35%。结论慢性乙型肝炎患者不同HBV特异性CTL表达KIR,这可能与慢性乙型肝炎患者HBV特异性CTL功能低下密切相关。
Objective To detect the expression of killer cell inhibitory receptor (KIR) on the surface of HBV-specific CTL cells in peripheral blood of chronic hepatitis B patients. Methods The expression of KIR on different HBV-specific CTLs in patients with chronic hepatitis B was detected directly by using pentamer and flow cytometry in MHC-Ⅰ-peptide-pentamer. Results In patients with chronic hepatitis B, KIR positive CD8 + T cells were significantly increased; KIR positive HBcAg (18-27) specific CTL, HBeAg (335-343) specific CTL, HBp (575-583) specific CTL The percentages were 12%, 20%, 35% respectively. Conclusions Different HBV-specific CTLs express KIR in patients with chronic hepatitis B, which may be closely related to HBV-specific CTL dysfunction in patients with chronic hepatitis B.
