目的:对大鼠离体心脏进行Langendoff灌流,观察油茶皂甙(SQS)对心肌缺氧/复氧(I/R)损伤的保护作用。方法:灌流开始后,将心脏随机分为4 组。Norm al组:有氧灌流;缺氧/复氧(I/R)组:缺氧40 m in 复氧30 m in;SQS大剂量组(SQSI):SQS05 m g/L预灌15 m in,作I/R模型;SQS小剂量组(SQSⅡ):SQS025 m g/L预灌15 m in,作I/R模型。记录心功能,测量心肌酶活性。结果:SQSⅠ、Ⅱ组与I/R组相比,SQS能增加心肌的收缩功能,提高心室最大收缩压(LVSP),左室内压最大变化速率(dp/dtmax),加快心率(HR);使冠脉流出液肌酸激酶(CK)生成减少;降低心肌组织钙含量,增强细胞膜Na+ K+ ATPase 及线粒体膜Ca2+ ATPase活性。结论:SQS具有抗钙超载及心肌保护作用。
OBJECTIVE: To perform Langendoff perfusion on isolated rat heart and observe the protective effect of Camellia saponin (SQS) on myocardial hypoxia/reoxygenation (I/R) injury. METHODS: After perfusion was started, hearts were randomly divided into 4 groups. Norm al group: aerobic perfusion; hypoxia/reoxygenation (I/R) group: hypoxia 40 m in reoxygenation 30 m in; SQS high-dose group (SQSI): SQS0 5 m g/L pre- irrigation 15 m In, for I/R model; SQS low dose group (SQSII): SQS025 m g/L pre-injection 15 m in for I/R model. Heart function was recorded and myocardial enzyme activity was measured. RESULTS: Compared with the I/R group, SQSI and SQS group increased myocardial systolic function, maximal ventricular systolic pressure (LVSP), maximum rate of left ventricular pressure (dp/dtmax), and accelerated heart rate (HR); Coronary effluent creatine kinase (CK) production decreased; reduced myocardial calcium content, enhanced membrane Na + K + ATPase and mitochondrial membrane Ca 2+ ATPase activity. Conclusion: SQS has anti-calcium overload and myocardial protection.
