本文提出了邻氨基酚 (OAP) -H2 O2 -辣根过氧化物酶 (HRP)伏安酶联免疫分析体系并用于烟草花叶病毒 (TMV)的测定。本方法以线性扫描二阶导数伏安法检测HRP催化H2 O2 氧化OAP的产物 ,用于HRP测定的检出限为 1 5× 1 0 - 1 mU·L- 1 ,线性范围为 2 5× 1 0 - 1 ~ 1 .0× 1 0 3mU·L- 1 。本法对TMV测定的检测限为 2 .0ng·mL- 1 。用所建立的方法测定病毒感染病叶澄清液的最高稀释比为 1∶3.9× 1 0 6
In this paper, an o-aminophenol (OAP) -H2 O2 - horseradish peroxidase (HRP) voltammetric enzyme-linked immunosorbent assay system was developed and applied to the determination of tobacco mosaic virus (TMV). In this method, the linear derivative of second order derivative voltammetry was used to detect the product of OAP oxidized by H2O2 catalyzed by H2O2. The detection limit of HRP for the determination of HRP was 15 × 10 ~ 1 mU · L-1 with a linear range of 25 × 1 0 - 1 ~ 1 .0 × 10 3 mU · L -1.
