目的建立同时测定重楼中重楼皂苷Ⅰ、Ⅱ、Ⅵ、Ⅶ的超高效液相色谱-蒸发光散射(UPLC-ELSD)分析方法。方法采用WatersAcquity UPLC H-Class色谱系统,ELSD检测器,BEH C18色谱柱(50 mm×2.1 mm,1.7μm),流动相为乙腈-水,以体积流量0.45 mL/min进行梯度洗脱,柱温30℃。结果被测成分在线性范围内均具有良好的线性关系(r>0.999 5);平均回收率在98.36%~100.11%,RSD≤1.56%。结论 UPLC法分离效果及重现性好,且快速、简便,可作为重楼的质量控制方法。
OBJECTIVE To establish a method for the simultaneous determination of Saponins Ⅰ, Ⅱ, Ⅵ and Ⅶ in Reiculovirus by UPLC-ELSD. Methods Waters Accity UPLC H-Class chromatography system, ELSD detector, BEH C18 column (50 mm × 2.1 mm, 1.7 μm) were used. The mobile phase consisted of acetonitrile-water and eluted with a flow rate of 0.45 mL / 30 ° C. Results The linear range of the tested components was linear (r> 0.999 5). The average recoveries ranged from 98.36% to 100.11% with RSD ≤ 1.56%. Conclusion The UPLC method has good separation and reproducibility, and is fast and simple. It can be used as a quality control method for re-building.
