目的采用吸附树脂与离子交换树脂联合去除人参提取物杂质,建立人参皂苷的纯化方法。方法人参醇提物不溶液经吸附树脂AB-8除去水溶性杂质后,再用强碱树脂D280进一步脱色纯化;并通过HPLC等多种分析手段,监控树脂纯化过程中各阶段的树脂除杂脱色能力以及皂苷的损失程度,验证本法的可靠性。结果树脂联用处理红参醇提物后,除杂和脱色能力由单用吸附树脂时的94.7%和93.0%分别提高到99.6%、97.4%,样品纯度由单用吸附树脂时的84.21%提高到98.57%,皂苷损失小于2%。结论本方法简便、快速、回收率高,可用于人参皂苷的大批量纯化。
Objective To use adsorption resin and ion exchange resin to remove impurities of ginseng extract and establish a method for purifying ginsenosides. Method The ginseng alcohol extract was not dissolved in the solution by AB-8 to remove water-soluble impurities, and then further decolored and purified by strong base resin D280. The resin removal and decolorization at each stage of the resin purification process was monitored by HPLC and other analytical methods. The ability and degree of loss of saponins verify the reliability of this method. RESULTS After the resin was treated with red ginseng alcohol extract, the impurity removal and decolorization capacity increased from 94.7% and 93.0% when the resin was used alone to 99.6% and 97.4%, respectively, and the sample purity increased from 84.21% when the resin was used alone. To 98.57%, saponin loss is less than 2%. Conclusion This method is simple, rapid, and has a high recovery rate. It can be used for the large-scale purification of ginsenosides.
