目的研究阳离子脂质体联合转铁蛋白(transferrin ,Tf)介导报告基因虫荧光素酶基因对人肿瘤细胞的靶向转染。方法比较阳离子脂质体Lipofectin、Lipofectin联合含双铁的人转铁蛋白[HTf(Fe) 2 ]介导质粒pDR2 luc转染人肝癌细胞株HepG2 、SMMC772 1、人肾癌细胞株GRC及非洲绿猴肾细胞COS7的转染效率,转染效率通过以液体闪烁计数仪单光子计数法测定荧光素酶活性估计。结果除COS7外,其余3株细胞以Lipofectin联合HTf(Fe) 2 介导pDR2 luc的转染效率均比单用Lipofectin的转染效率高(P <0 0 1) ,其中HepG2 与SMMC772 1中的转染效率增高8倍多,GRC中的转染效率增高4倍多。结论Lipofectin联合HTf(Fe) 2 介导质粒pDR2 luc转染细胞时对人肿瘤细胸株有一定的靶向作用。
Objective To study the targeted transfection of reporter gene luciferase gene into human tumor cells by cationic liposomes combined with transferrin (Tf). METHODS: Lipofectin and Lipofectin were transfected into human hepatocellular carcinoma cell lines HepG2, SMMC772 1, human renal cell carcinoma cell line GRC and African green cells mediated by double iron-mediated human transferrin [HTf (Fe) 2] Transfection efficiency of monkey kidney COS7, transfection efficiency was estimated by measuring luciferase activity by liquid scintillation counting single photon counting. Results The transfection efficiency of pDR2 luc mediated by Lipofectin combined with HTf (Fe) 2 was higher than that transfected with Lipofectin alone (P <0.01) except for COS7. The transfection efficiency of HepG2 and SMMC772 1 Transfection efficiency increased more than 8 times, GRC transfection efficiency increased 4 times. Conclusion Lipofectin combined with HTf (Fe) 2-mediated plasmid pDR2 luc transfected cells have certain targeting effect on human breast cancer plant.
