对大鼠骨髓间充质干细胞(BMSCs)分离培养、体外扩增,采用含不同葡萄糖浓度的培养基加尼克酰胺、exendin-4向胰岛样细胞进行诱导。将诱导细胞通过尾静脉及肾包囊移植入糖尿病大鼠模型体内,通过对血糖的检测,观察诱导后细胞对糖尿病大鼠模型的治疗效果。发现大鼠BMSCs在尼克酰胺和exendin-4作用后,可通过尾静脉和肾包囊途径进入体内,分布在不同脏器,同时产生降低链脲佐菌素(STZ)诱导糖尿病大鼠血糖水平的作用。
Rat bone marrow-derived mesenchymal stem cells (BMSCs) were isolated and cultured, expanded in vitro, and cultured in pancreatic islet-like cells with different concentrations of glucose, such as gannamide and exendin-4. The induced cells were transplanted into the diabetic rat model through the tail vein and the renal capsule. The blood glucose was measured to observe the therapeutic effect of the induced cells on the diabetic rat model. After BMSCs were treated with nicotinamide and exendin-4, BMSCs of rats were found to enter the body through the tail vein and nephrotoxicity, distribute in different organs and reduce the blood glucose level in STZ-induced diabetic rats effect.
