本文采用阴离子交换剂DEAE-纤维素柱层析的方法,从接受MTX的家免(100 mg/kg)和肿瘤病人(总剂量2 g)的生物液中,分离MTX和它的主要代谢产物7-OH-MTX,并分别于302 nm、305 nm处测定其吸收度,该法灵敏度1×10~(-7)M,回收率93.6%,而且克服了其它方法,如全放射学分析、荧光分析、放射免疫、酶抑制等不能区别MTX、7-OH-MTX和叶酸同系物等的缺点,为临床药理的研究提供了有效的实验依据。
In this work, MTX and its major metabolite, 7, were isolated from biological fluids of home-based MTX-free (100 mg / kg) and tumor patients (total dose of 2 g) using anion exchange chromatography on a DEAE- -OH-MTX. The absorbance was measured at 302 nm and 305 nm, respectively. The sensitivity of this method was 1 × 10 -7 M, the recovery rate was 93.6%. Other methods such as total radiometric analysis, fluorescence Analysis, radioimmunoassay, enzyme inhibition and other shortcomings that can not distinguish MTX, 7-OH-MTX and folic acid homologues provide an effective experimental basis for the study of clinical pharmacology.
