目的 :研究两种不同的抗癌药物诱导细胞凋亡的规律 ,为卵巢癌的化疗提供理论依据。方法 :紫衫醇与顺铂分别作用于体外培养的卵巢癌SKOV3 细胞系 ,通过组织细胞化学染色、荧光染色、电子显微镜、DNA凝胶电泳、流式细胞术等方法进行检测和观察。结果 :经两种抗癌药物处理的卵巢癌SKOV3 细胞出现典型的细胞凋亡特征 ,在顺铂处理 0~ 36小时内 ,细胞凋亡呈均一的持续性增强 ,在 5~ 2 5nmol ml剂量范围内呈依赖性增强 ,凋亡率最高达 72 %。紫衫醇诱导细胞凋亡亦具有时间和剂量依赖性 ,2 5nmol ml浓度处理 12小时 ,凋亡率最高达 5 5 % ;但时间过长 (>12小时 )或浓度过大 (>5 0nmol ml) ,凋亡率下降。结论 :紫衫醇与顺铂能有效地诱导肿瘤细胞凋亡 ,诱导细胞凋亡是化疗药物治疗癌症的重要机制之一
Objective : To study the rule of apoptosis induced by two different anticancer drugs and provide theoretical basis for ovarian cancer chemotherapy. METHODS: Paclitaxel and cisplatin were applied to cultured ovarian cancer SKOV3 cell lines, respectively, and detected and observed by histochemical staining, fluorescent staining, electron microscopy, DNA gel electrophoresis, and flow cytometry. RESULTS: Typical ovarian epithelial cell characteristics were observed in two ovarian cancer SKOV3 cells treated with two anticancer drugs. Apoptosis was consistently and consistently increased within 0 to 36 hours of cisplatin treatment, ranging from 5 to 25 nmol. The intracellular dependence was enhanced and the apoptotic rate was up to 72%. Paclitaxel-induced apoptosis was also time- and dose-dependent, with 25 nmol ml treatment for 12 h, and the apoptotic rate was up to 55 %; however, the time was too long (>12 h) or the concentration was too large (>50 nmol ml ), the rate of apoptosis decreased. Conclusion :Effectiveness of paclitaxel and cisplatin on tumor cell apoptosis is one of the most important mechanisms for cancer treatment by chemotherapy drugs.
