目的探讨射线诱发肾癌细胞凋亡的作用和射线对肾癌细胞P53,Bcl-2基因表达的影响。方法利用末端脱氧核苷酰转移酶标记的流式细胞技术定性和定量分析射线诱导肾癌细胞凋亡。利用P53和Bcl-2单抗进行SPTM法免疫组织化学染色。结果0Gy、5Gy、10Gy、15Gy和20Gy照射后细胞凋亡量分别为1.02%、0.03%、7.03%、4.22%和36.09%。0Gy照射组P53表达标记指数为0.80%,20Gy照射后P53表达明显增加达23.80%。0Gy照射组Bcl-2基因表达为24.40%,20Gy照射后Bcl-2基因表达明显减少为1.80%。结论低剂量X射线照射诱发肾癌细胞凋亡量很低,20Gy时明显增加。射线诱发肾癌细胞凋亡伴随P53基因表达增加和Bcl-2基因表达减少,提示射线诱发细胞凋亡是多基因相互影响,相互作用的结果。
Objective To investigate the effect of radiation on the apoptosis of renal cell carcinoma and the effect of radiation on the expression of P53 and Bcl-2 in renal cell carcinoma. Methods Flow cytometry using terminal deoxynucleotidyl transferase was used to characterize and quantify the radiation-induced apoptosis of renal cell carcinoma. SPTM immunohistochemical staining using P53 and Bcl-2 mAb. Results The apoptotic rates were 1.02%, 0.03%, 7.03%, 4.22% and 36.09% after 0Gy, 5Gy, 10Gy, 15Gy and 20Gy irradiation, respectively. The expression index of P53 in 0Gy irradiation group was 0.80%, and the expression of P53 increased significantly by 23.80% after 20Gy irradiation. The expression of Bcl-2 gene in the group of 0Gy irradiation was 24.40%, and the expression of Bcl-2 gene in the group of 20Gy irradiation was significantly decreased to 1.80%. Conclusion The apoptosis of renal cancer cells induced by low-dose X-ray irradiation is very low and significantly increased at 20 Gy.
