目的探讨GPC对人胰腺癌细胞株BXPC-3体外生长抑制作用及机制。方法经预实验选用不同浓度GPC分别作用于BXPC-3细胞24h、48h后采用MTT法检测不同浓度GPC对BXPC-3细胞增殖的影响;免疫组织化学染色SP法检测GPC作用后COX-2的影响;FCM法检测GPC对细胞周期的影响。结果 GPC以时间剂量关系抑制BXPC-3的增殖并下调COX-2的表达;诱导细胞停滞在G0/G1期。结论 GPC可在体外明显抑制BXPC-3细胞的增殖,阻滞细胞周期,诱导细胞凋亡,而这种作用可能与COX-2的表达有关。
Objective To investigate the inhibitory effect of GPC on human pancreatic cancer cell line BXPC-3 in vitro and its mechanism. METHODS: BXPC-3 cells were treated with GPC at different concentrations for 24 h and 48 h respectively. The effects of different concentrations of GPC on the proliferation of BXPC-3 cells were detected by MTT assay. The effect of COX-2 after GPC was detected by immunohistochemistry The influence of GPC on cell cycle was detected by FCM. Results GPC dose-dependent inhibition of BXPC-3 proliferation and down-regulation of COX-2 expression; induced cell arrest in the G0 / G1 phase. Conclusion GPC can significantly inhibit the proliferation of BXPC-3 cells in vitro, arrest the cell cycle and induce apoptosis, which may be related to the expression of COX-2.
