采用中和活性法、免疫印迹法 ,通过α1b、α2b、α2a干扰素与自制的 6株单抗反应的结果差异 ,对 3种干扰素加以鉴别。中和活性结果表明 ,AK1单抗只与IFN α1b有中和活性 ,AK2单抗只与IFN α2有中和活性 ,而AK4单抗与IFN α1b和IFN α2b皆有中和活性。免疫印迹结果表明 ,AK1单抗只与IFN α1b有印迹反应 ,AK2与IFN α2有印迹反应 ,而AK4单抗与IFN α1b和IFN α2b皆有印迹反应 ,与IFN 2a无印迹反应。AK1或AK2能够鉴别IFN α1b与IFN α2 ,AK4能够鉴别IFN α2a与IFN α2b。
Neutralization activity and immunoblotting were used to identify three kinds of interferon through the differences in the results of the reaction of α1b, α2b and α2a interferon with the six homemade monoclonal antibodies. Neutralizing activity results showed that AK1 monoclonal antibody only had neutralizing activity with IFNα1b, AK2 monoclonal antibody only had neutralizing activity with IFNα2, and AK4 monoclonal antibody had neutralizing activity with IFNα1b and IFNα2b. Immunoblotting results showed that AK1 McAb only had the imprinting reaction with IFNα1b, the imprinting reaction of AK2 and IFNα2, but the imprinting reaction of AK4 monoclonal antibody with IFNα1b and IFNα2b, and no imprinting reaction with IFN 2a. AK1 or AK2 were able to identify IFNα1b and IFNα2, AK4 was able to identify IFNα2a and IFNα2b.
